Abstract

A study on the immobilisation of peroxidase enzyme in sol-gel-chitosan hybrid monolith for the preparation of hydrogen peroxide optical transducer has been conducted for biosensor fabrication. This research involved the optimisation of the sol-gel-chitosan ratio, characterization of enzyme in free solution and immobilization of peroxidase enzyme in sol-gel-chitosan hybrid. The characterizations were studied by using UV-Visible Spectrophotometer. The results showed that the enzyme did not change its biological activities after immobilisation. The optimised ratio of sol-gel:chitosan was 4:6. The relative standard deviation (RSD) values for the reproducibility of the method based on free and immobilized enzyme were 2.71 % and 3.76%, respectively.
