Detection of trophozoites surface proteins (tsaII) gene of Giardia duodenalis obtained from Malaysian Aborigines using polymerase chain reaction (PCR)

Abstract

Objective: To detect trophozoites surface protein (tsaII) gene of Giardia duodenalis obtained from Malaysian Aborigines using polymerase chain reaction (PCR). Material and Methods: The study was carried out on six Malaysian isolates of Giardia duodenalis (110, 7304, 6304, M007, 2002 and 6307) from faeces of aborigine patients admitted to Gombak Hospital (Kuala Lumpur). The WB was a reference pathogenic strain from human and G. muris was from wild mouse. Both strains were commercially obtained from the American Type Culture Collection (ATCC). All the isolates were cultured axenically in TYI-S-33 medium. One sets of primers were used for the techniques; primers (LP3 and RP3). Results: The sets of primers amplified trophozoites surface proteins (tsaII) gene of 520 bp in size. The study showed that the sets of primers could detect G. duodenalis isolates from the patients specifically. Conclusion: The sets of primers (LP3 and RP3) are specific and sensitive in the detection of trophozoites surface proteins (tsaII) gene of Giardia duodenalis.
