Apoptosis induction in CEM-SS T-lymphoblastic leukemic cell line by goniothalamin

Abstract

Goniothalamin is a biologically active plant styrylpyrone derivative isolated from Goniothalamus sp. In this study, the cytotoxic effects and the mode of cell death induce by goniothalamin in T-lymphoblastic leukemia cell line (CEM-SS) were determined. The 72 hours cytotoxic dose fifty percent (CD50) value of goniothalamin in CEM-SS cells was 0.9 μg/ml, which was comparable to etoposide (0.6 μg/ml), a drug which is known to induce apoptosis in many cell lines. Goniothalamin induced apoptosis was detected as early as two hours after treatment and reached up to 66% at 72 hours which was higher than the induction by etoposide. Various stages of apoptotic cells and the presence of apoptotic bodies, in addition to internucleosomal DNA fragmentation, confirmed the apoptotic death in goniothalamin treated cells.
