Abstract

This study was carried out to identify the biological activity of aqueous extracts of four plants manjakani (Croton caudatus), tongkat Ali (Eurycoma longifolia), haji Samat (Prismatomeris glabra) and kerbau amuk (Schefflera heterophylla) specifically on the antibacterial activity, cytotoxicity towards Vero cells and antiviral activity against herpes simplex virus type-1 (HSV-1). Only P. glabra showed low bacteriocidal activity towards all tested bacteria. Exposure towards Vero cell lines indicates the concentration that causes 50% death of the population of cells (CC50) for C. caudatus, E. longifolia, P. glabra and S. heterophylla were 0:22, 0:28, 0:25, 0:40 mg/mL respectively indicating the non-cytotoxicity of all the extracts. With no cytotoxicity effect, the antiviral activity was determined against HSV-1 with the ability to reduce 50% of plaque formation concentration values (EC50) were 0.06, 0.03, 0.7 and 0.062 mg/mL against C. caudatus, E. longifolia, P. glabra and S. heterophylla respectively. In order to determine the effectiveness of extract as antiviral agent, the selectivity index (SI) for C. caudatus, E. longifolia, P. glabra and S. heterophylla was calculated as 3.7, 9.3, 3.6 and 6.0 respectively. Based on these values, all extract has moderate potential as anti-HSV-1. In conclusion, all aqueous extract of four plants tested had low bacteriocidal and moderate antivirus ability with the highest selectivity value shown by E. longifolia. The low cytotoxicity value shown in this study implied non-cytotoxic effect of all the extracts.
