A study on chitosan macroparticles as potential affinity beads

Abstract

The chitosan macroparticles were prepared using precipitation method where macroparticles with average diameter, d v of 2-mm diameter prepared through 31-gauge needle was chosen to investigate its potential as affinity beads. The amine groups on the chitosan macroparticles were detected using 2,4,6-trinitrobenzenesulfonic acid (TNBS) assay where orange colored derivative compound was observed. Furthermore, the amino group concentration on the chitosan macroparticles was determined using a developed glycine standard curve, which was 0.41 ± 15.32-μg per mg chitosan. The macroparticles were then functionalized with lactose via reduction amination for 48 hours at room temperature. Using Fourier-transform infrared (FTIR) spectroscopy, additional peaks were observed for the lactose-functionalized chitosan macroparticle, which were at wavenumber 1028.56 cm -1 and 2900 cm -1 , indicating successful modification and show chitosan macroparticles can be functionalized and further use as affinity beads.
