A potentiometric biosensor based on plasticiser free methacrylic-acrylic membranes with immobilized AChE for the evaluation of pesticides toxicity

Abstract

An enzyme based toxicity biosensor using methacrylic-acrylic membrane, which is based on the principle of irreversible inhibition caused by pesticides, has been developed in this work. A potentiometric enzyme biosensor was constructed from acetylcholinesterase (AChE) to measure loss of enzyme activity in the presence of pesticide inhibitors. From the study, the pH selective sensor gives near Nernstian response of about 55 mV/decade while the respond from the enzyme layer is about 28 mV/decade change of substrate concentration. 71% inhibition was achieved with a 1 ppm solution of dichlorvos and at 0.5 ppm the inhibition was about 58%.
